In situ hybridization for P-selectin, leukocyte-specific antigen-I, Mac-1, and CD18 on capillary-like structures in the developing rat CNS. The endothelial cells of the cerebral microvasculature of the rat brain were examined for the expression of P-selectin, a cell surface protein that is a ligand for selectins, and for the expression of leukocyte-specific antigen-I (LSA-I) and beta 2 integrin family members (Mac-1 and CD18). P-selectin was detectable from embryonic day (ED) 10 and was abundant on microvessels from ED 12 to 17. However, a few P-selectin positive microvessels were still seen in the brain on ED 18. On ED 13, LSA-I, Mac-1, and CD18 were expressed on blood vessels in the brain, including microvessels. These molecules were present in microvessels from ED 15 onward. In situ hybridization demonstrated the presence of messenger RNA (mRNA) for these molecules in the endothelial cells of microvessels from ED 13 to 17. The expression of P-selectin on blood vessels in the brain was time dependent and preceded that of Mac-1 and CD18.
